AAV Titer Estimation With Mass Photometry

Explore the fast, efficient, and accurate nature of mass photometry for estimating AAV titer. This application note reviews the performance of the SamuxMP line of mass photometers, featuring a new titer estimation tool, which provides titer data on all populations in the sample. A study conducted using SamuxMP Auto addresses the importance of measuring capsid titer in evaluating the quality of AAV samples and introduces mass photometry as a powerful analysis technique for this purpose. Mass photometry can measure the mass of individual AAVs in solution and provide information about critical analytical attributes of AAV samples. The study found that mass photometry is a reproducible and comparable method for estimating AAV titer, with results in agreement with ELISA measurements. The study also evaluated the use of mass photometry for measuring AAV titer in clone screening and different purification stages, finding good agreement with ELISA measurements and comparable results to dPCR and Unchained Labs' Stunner.
